The Dpg Gene : An Intracorpuscular Modifer of Red Cell Metabolism
نویسنده
چکیده
The genetic locus designated Dpg has two alleles in outbred Long-Evans rats. Genotype at this locus affects quantities of red cell 2.3diphosphoglycerate (DPG) and adenosine triphosphate, as well as activities of two important glycolytic enzymes: phosphofructokinase and pyruvate kinase. Intravascular red cell survival is shortened in low-DPG animals. In order to get closer to the specific action of this locus. we addressed the question of whether the Dpg gene acts through intracorpuscular or extracorpuscular factors. Bone marrow transplantation after total body irradiation and 51Cr red cell survival after cross transfusion were the methods used. Because the animals

The Dpg gene: an intracorpuscular modifier of red cell metabolism.
The genetic locus designated Dpg has two alleles in outbred Long-Evans rats. Genotype at this locus affects quantities of red cell 2,3- diphosphoglycerate (DPG) and adenosine triphosphate, as well as activities of two important glycolytic enzymes: phosphofructokinase and pyruvate kinase. Intravascular red cell survival is shortened in low-DPG animals. In order to get closer to the specific acti...

Red cell carbonic anhydrase activity, 2.3 DPG concentration, and activities of three key enzymes controlling DPG metabolism (PK, PFK and DPGM) were measured in normoxic and hypoxic (incubation in 13.5% O2) chick embryos. In normoxia 2.3 DPG concentration and carbonic anhydrase activity begin to increase by the third week of incubation. Hypoxia induces a rise of 2.3 DPG concentration and carboni...
متن کاملThe effects of deoxygenation of adult and fetal hemoglobin on the synthesis of red cell 2,3-diphosphoglycerate and its in vivo consequences.
Patients over 1 month of age with arterial oxygen pressures of less than 60 mm Hg were found to have elevated red cell 2,3-diphosphoglycerate (2,3-DPG) levels and blood with a decreased affinity for oxygen. The increase in 2,3-DPG was proportional to the degree of hypoxemia. In patients under 1 month of age this relationship was not observed. Red cells from adults, but not newborns, showed rapi...
متن کاملThe relationship between red cell 2,3-diphosphoglycerate and levels of hemoglobin in the human.
The central role of 2,3-diphosphoglycerate (DPG) in the metabolism and function of most mammalian red cells is only beginning to become apparent. 2,3-Diphosphoglycerate, a glycolytic intermediate present in high concentration only in the red cell, has been said to function as a storage source for phosphate.1 It may also act as a metabolic regulator, since DPG has been found to exert significant...
متن کاملPyruvate kinase hyperactivity genetically determined metabolic consequences and molecular characterization.
Four people from the same family with red cell pyruvate kinase (PK) hyperactivity are described. These people displayed low 2,3-diphosphoglycerate (2,3-DPG) and high adenosine triphosphate (ATP) levels. In vitro metabolism studies of their red cells showed the relationship between the PK activity, low 2,3-DPG, and high ATP levels. The PK electrophoretic pattern of these subjects was abnormal by...
